Well I have a DS lite with an R4 and I want to upgrade to a DSi at the local Gamestop. The question is, are you able to use the R4 cartridge with the AceKard 2i software downloaded from the website?

Wait... you want to put firmware from one flash cart onto a different flash cart and hope it works? haha

    no

firmware is specific to the card,

if you want to upgrade to dsi you need to upgrade your card too

Yah, unfortunately the firmware for the AceKard does not work for the R4 cart. I tried it; didn't work. poop. lol.

Mmmh, it sounds tempting to jump on to the DSi with the AceKard. My left trigger is worn out on my DS lite too... I could be wrong, but doesn't the DSi have an update firmware from time to time? Eventually the AceKard would be booted off right? Assuming you kept up with the DSi firmware that is... That's one thing that's keeping me from upgrading to a DSi and getting an AceKard.

Yes, which is why the teams such as Acekard suggest not update. A new flashcart such as SuperCard DSONEi should be able to get around these 'blocks' however, with their new methods of firmware flashing.

Sorry, I do not think that you can...but can I ask why you would want to trade your lite for a DSi? The DSi have one of the worst battery life's in all of the handheld consoles, they do not offer GBA compatibility at the moment (they could show up as a download on the DS online shop, wouldn't get ones hopes up), and the cameras are only .3 MP (no practical use for that).
